摘  要:采用pH梯度法制备了阿霉素脂质体.用SephadexG-25微型柱分离-紫外分光光度法测定阿霉素脂质体包封率.结果表明,当药物/磷脂重量比为0.02~0.05时,包封率均大于90%;体外泄漏研究显示,25℃时,2h内泄漏百分率约为2.7%,保证了临床应用.Adriamycin liposomes(ADML) were prepared by a pH gradient method and the encapsulation efficiency was determined spectrophotometrically by UV absorbance at 480 nm after the free drug molecules was seperated from ADML on a sephadex G 25 minicolumn.The results obtained indicated that the encapsulation efficiency was over 90% when the drug/phospholipid weight ratios were between 0 02 and 0 05.The extent of leakage from liposome was examined by incubation of ADML with 0 9% NaCl at 25℃.The results showed that approximately 2 7% of the entrapped drug was released from ADML within 2 h.
