早期自然流产患者绒毛Notch受体mRNA和蛋白表达及与妊娠关系的初步探讨

摘　　要：目的:初步探讨早期自然流产患者绒毛Notch受体(Notch1、Notch2、Notch3、Notch4)mRNA及蛋白表达及对滋养细胞的调控作用。方法:用实时荧光定量逆转录聚合酶链反应技术和免疫印迹法检测6例自然难免流产患者和12例正常早期妊娠妇女绒毛Notch1、Notch2、Notch3、Notch4的mRNA和蛋白的表达水平。结果:(1)自然流产组绒毛Notch1和Notch3 mRNA表达水平高于正常早期妊娠组(P<0.05);(2)Western-blot检测显示早期自然流产患者绒毛中Notch1、Notch2、Notch3受体蛋白的表达水平高于正常早期妊娠组(P<0.05)。结论:Notch受体蛋白在早期自然流产患者绒毛中表达上调,可能与自然流产的发病原因有关。Objective：To investigate the mRNA and protein expression of Notch receptors（ Notch1, Notch2, Notch3, Notch4） in villi of first trimester spontaneous abortion and its role in regulating trophoblasts. Methods：Real-time fluorescence quantitative RT-PCR and Westernblot were used to detect the Notch receptors mRNA and protein expression level,villi was obtained from 6 women with first spontaneous abortion and 12 normal first trimester pregnant women. Results： （ 1 ） Expression of Notch1 mRNA and Notch3 InRNA was significantly up-regulated in villi of first trimester spontaneous abortion compared to normal pregnancy（P 〈0.05）. （2） Western-blot analysis demonstrated that expression of Notch1, Notch2 and Notch3 receptor protein in villi of first trimester spontaneous abortion was significantly up-regulated in comparison to normal pregnancy （ P 〈 0.05 ）. Conclusion ： The up-regulated expression of Notch receptor protein in villi of first trimester spontaneous abortion may be related with the cause of the first trimester spontaneous abortion.
