摘 要:目的:通过部分肝切除建立小鼠肝再生动物模型,研究肝脏再生过程中形态和血清学变化趋势,为研究肝再生调控机制奠定实验基础。方法:采用50%肝切除方法建立C57/BL6小鼠肝再生模型;分别于手术后第1、第5、第9、第13、第21和第36天,摘眼球法取血,分离血清后检测ALT、AST、AFP和ALB,并进行C-反应蛋白(CRP)、肝细胞生长因子(HGF)、表皮生长因子(EGF)、转化生长因子-α(TGF-α)等细胞因子的检测。并取肝脏组织进行苏木素-伊红(HE)染色,研究肝再生过程中形态学改变。结果:光镜下可见肝切除后肝细胞核分裂极其活跃;ALB分泌降低,后逐渐升高至正常值;ALT、AST先升高后逐渐下降,但一直高于正常值;AFP持续为阴性;CRP在术后5d内迅速升高,之后有所下降;HGF在术后持续保持较高水平,且在死亡小鼠中也有测到高值的HGF;EGF与TGF-α都升高。结论:多种细胞因子参与肝再生调控,在肝再生过程中,能够继续维持白蛋白合成、有毒物质降解等肝细胞特异性功能而保持机体的稳定。Objective; To study the changes in morphology and serum during the process of liver regeneration, which may lay the experimental foundation of the research of the accommodation mechanism of liver regeneration. Nethods: Fstablish a C57/BL6 mice model of liver regeneration by 50% hepatectomy. After surgery, take the blood samples of mice on 1st, 5th, 9th, 13th, 21th, 36th, days, measure the vaiue of ALT, AST, AFP, ALB, and detect the cytokines of C-reactive protein (CRP), liver cell growth factor (HGF), epidermal growth factor (EGF) and transforming growth factor-α (TGF-α ). And then observe the morphological changes of liver ceils during the regeneration process by Hematoxylin and Eosin Staining. ResuIts: The multiplicative division of nucleus of Liver ceils was extremely active under the light microscopy after the surgery. The production of Albumin (ALB) was depressed after the surgery, and increased gradually to normal level on the 36th day. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were increased rapidly just after the surgery, and then decreased gradually to the normai level. AFP was all negative during the research time. The degree of the creactive protein(CRP) extremely rose in the first five days, and then it decreased a little, but stili above the normal value. Hepatocyte growth factor (HGF) sustained increase, which could also be detected in the died mice. Fpidermal growth factor (HGF) and Transforming growth factor-α (TGF-α ) were both increased. Conclusion: A variety of cytokines are involved in the liver regeneration accommodation, In the regeneration process, liver can still remain some liver-specific functions, such as albumin synthesis, toxin degradation and so on. thereby can maintain the stability of the body.
