摘 要:目的:观察升压宁对内毒素休克肺损伤大鼠肺组织中肿瘤坏死因子-α(TNF-α)和诱导型一氧化氮合酶(iNOS)的影响。方法:采用一次性静脉注射内毒素(LPS)10 mg/Kg复制内毒素休克模型,以升压宁预处理动物,并以地塞米松为阳性对照,HE染色光镜下观察肺脏的组织形态学变化;免疫组化法测定肺组织中TNF-α和iNOS的表达。结果:内毒素休克模型组存在严重肺损伤,肺组织中TNF-α和iNOS表达较空白对照组显著增强(P<0.01),升压宁组肺损伤较模型组减轻,TNF-α和iNOS的表达与模型组比较显著降低(P<0.01)。结论:升压宁能减轻内毒素休克肺损伤,其机理与抑制TNF-α和iNOS的表达有关。Aims: To explore the effect of Sheng Ya Ling on TNF - α and iNOS in lung injury rats with endotoxie shock. Methods: The rars were injected intravenously LPS 10mg/Kg to make endotoxic shock models. The rats were pre - treated by Sheng Ya Ling with the rats given dexamethasone as positive control group. Then histological changes of lungs of both groups were observed with HE staining and the expression of TNF - α and iNOS were measured by immunohistochemical method. ResuLts: In the endotoxic shock model group, the lung of rats were severely injured and the expression of TNF - α and iNOS in lungs were increased more significantly than the blank control group ( P 〈 0.01 ) ; The lung injury of Sheng Ya Ling group was slighter than that of model group and expression of TNF - α and iNOS were decreased more significantly than that of model group ( P 〈 0. 01 ) . ResuLts: Sheng Ya Ling can reduce lung injury by endotoxic and its mechanism may be with inhabitation of expression of TNF -α and iNOS.
