摘 要:目的:探求不同产地和同一产地不同采收期的满山红油的高效液相(HPLC)特征色谱,为建立满山红油的色谱特征图谱提供依据。方法:按中国药典2005年版一部附录XD中挥发油测定的甲法,水蒸气蒸馏提取挥发油。采用梯度洗脱技术进行HPLC分离分析。色谱柱为Zorbax Eclipse XDB-C18柱,流动相为乙腈-0.1%磷酸水溶液,流速为1mL·min^-1;检测波长为210nm。用聚类分析方法处理色谱指纹信息,比较色谱以牻牛儿酮为参照色谱峰的32个特征峰的信息、单波长的全部指纹信息及200-350nm全波长的二维特征图谱的指纹信息。结果:所用的特征图谱测定系统对满山红油的分离良好,牻牛儿酮的纯色谱峰得到确认。采用200-350nm全波长的二维特征图谱的指纹信息较采用单波长全部特征指纹信息及色谱特征峰指纹信息的聚类结果更加可靠。结论:不同产地的满山红油中吉马酮的含量差异较大。不同产地的满山红油的色谱特征色谱比较相近,但有差别。200-350nm全波长的二维特征图谱的指纹信息更能全面表达满山红油的内在质量。OBJECTIVE To investigate HPLC profiles of folium rhododendri daurici oil from the different sources and the same sources of different harvesting seasons and provide basis for the establishment of chromatographic fingerprint of folium rhododendri daurici oil. METHODS According to the test for volatile oil method of appendix X D in ChP 2005, and the volatile oil is distillated by steam distillation. The HPLC was run on Zorbax Eclipse XDB-C18 column with acetonitrile-water (0. 1% phosphoric acid) as mobile phase in gradient elution at a flow rate of 1.0 mL·min^-1. The detection wavelength was 210 nm, respectively. The whole information of single-wavelength chromatographic fingerprint, two-dimension information data at full wavelength 200 - 350 nm and the date of 32 fingerprint peaks compared with Germacrone were treated by cluster analysis. RESULTS The HPLC fingerprint of foliurn rhododendri daurici oil were obtained with good resolution under the experiment chromatographic system. The pure peak of chromatographic fingerprints of Germacrone was identified. Better cluster result can be obtained from two-dimension information data at full wavelength 200 - 350 nm than the whole information of single-wavelength chromatographic fingerprint and the chromatographic characteristic peaks information. CONCLUSION There were greater difference on content of folium rhododendri daurici oil from different source. Though HPLC profiles in folium rhododendri daurici oil from the different sources are similarity, the difference of HPLC profiles can be founded. The fingerprints characterized by two-dimension information data at full wavelength could provide more overall information for the inherent quality of folium rhododendri daurici oil.
