摘 要:在pH=3.3的Britton-Robinson(B-R)缓冲溶液中,用循环伏安法研究了亚甲蓝(MB)与肝素(Hep)的相互作用.在最佳实验条件下,MB与Hep通过静电作用形成复合物,导致溶液的氧化峰电流和还原峰电流均明显降低,其峰电流差值(Δip)与Hep的浓度成正比,据此建立了一种测定Hep浓度的新方法,测定的线性范围为1.0~5.0μg/mL,相关系数R=0.9986,检测限(3σ)为0.23μg/mL.该法应用于注射液中肝素含量的测定,结果满意.The interaction between methylene blue (MB) and heparin (Hep) was studied by cyclic voltammerry in a pH 3.3 Britton-Robinson buffer solution. Under the optimum conditions, MB reacted with Hep to form a complex compound by virtue of electrostatic force, which resulted in significant decreases in both oxidation peak current and reduction peak current. The difference value of the peak currents (Aip) was proportional to the concentration of Hep in a certain range. Based on this result, a new method for the determination of Hep concentration was established. Its linear range for Hep determination was 1.0-5.0 g/mL, with a correlation coefficient of 0. 998 6 and a determination limit of 0. 23 g/mL. This method was applied to the determination of Hep in drug samples and satisfactory results were obtained.
