摘 要:目的观察麝香保心丸对脑缺血再灌注损伤大鼠Bcl-2、Caspase-3表达的影响及其是否存在剂量依赖性。方法采用双侧颈总动脉阻断的方法建立大鼠脑缺血再灌注损伤模型,缺血再灌注模型大鼠随机分为模型组、麝香保心丸高剂量组、麝香保心丸低剂量组,并予相应处理。采用连续切片免疫组化技术检测脑组织中Bcl-2、Caspase-3表达的部位及数量。结果麝香保心丸高剂量组及低剂量组皮层、纹状体Bcl-2免疫阳性细胞数与模型组相比差异明显,在再灌注后72、120、168h增加;麝香保心丸高剂量组及低剂量组皮层、纹状体Caspase-3免疫阳性细胞数与模型组比较差异明显,在再灌注后72h显著减少;同时麝香保心丸高剂量组皮层、纹状体Bcl-2、Caspase-3免疫阳性细胞数与低剂量组比较差异显著。结论麝香保心丸可以上调大鼠脑缺血再灌注损伤后脑组织皮层和纹状体Bcl-2的表达、下调Caspase-3的表达,从而起到一定的脑保护作用,并且具有剂量依赖性。Objective: To determine the effects of heart - protecting musk pill (HMP) on expression of Bcl - 2, Caspase- 3 after cerebral ischemia reperfusion injury in rats and the influence whether or not shows a dose-depended manner. Methods: By bilateral carotid artery occlusion method rat cerebral ischemia-reperfusion injury model, ischemia- reperfusion model rats were randomly divided into the model group, HMP high- dose treatment group, HMP low - dose treatment group. A continuous sections immunohistochemical detection of brain tissue in the Bcl - 2 and Caspase - 3 expression of the location and quantity. Results: HMP high - dose treatment group and low dose group cortex, striatum Bcl -2 immunoreactive cells compared with the model group there were significant differences, in 72,120,168h after reperfusion be increased ; HMP high - dose treatment group and low dose group cortex, Caspase - 3 striatum immunoreactive cells compared with the model group had a significant difference, in the 72h after reperfusion be reduced significantly; At the same time HMP high dose group cortex, striatum Bcl -2,Caspase - 3 immunoreactive cells with lowdose group had a significant difference. Conclusion: HMP can be raised rat cerebral ischemia - reperfusion injury cerebral cortex and striatum of Bcl - 2 expression, reduced the expression of Caspase - 3, which plays a role in brain protection, and has a dose - dependent manner.
