摘 要:【研究目的】为了利用rDNA ITS区的遗传变异的特点,为镰刀菌种内及种间的分类鉴定提供一定的分子技术辅助手段,【方法】试验以PMD18-T为载体,对10个不同镰刀菌菌株的rDNA的ITS区及28S部分片段进行了克隆、测序,用DNAMAN4.0软件进行序列分析,分析了菌株间的同源性及遗传距离,并建立了分子系统进化树。【结果】试验结果表明:所测各菌种间的亲缘关系在92.2%-99.9%之间,反映了种间较大的遗传差异,这与形态学上把它们鉴定为不同的种相吻合,而从不同地区、不同寄主上分离的同一菌种的不同专化型,其同源性高达96.6%-100%,说明同一菌种的不同专化型,在rDNA基因序列上碱基差异甚微,同源性很高。【结论】利用ITS对镰刀菌进行种间的分类鉴定是稳定可行的,但对于种内及专化型的鉴定存在困难。[OBJECTIVE]In order to take the advantage of genetic variation of rDNA ITS areas of Fusarium and providing a means for the identification of the different kind species and the different specialized form,[METHOD]This experiment take the PMD18-T as the vector, expanding and sequencing the ITS and 28S partial sequences of ten Fusarium strains, utilizing the DNAMAN4.0 software to analyze the multiple sequence and the genetic distance, establishing the molecular system evolution tree. [RESULTS] The result indicates that the genetic relationship of each strains between the 92.2% to 99.9%, reflect the inter-species have bigger genetic diversity, This is coincide with the different kind species which is appraised by the morphology analysis, but the same species which are different specialized types and biological races from the different area or host which have the higher homology about between 96.6% to100%. It is possible that biological races are higher homology, the difference is very small. [CONCLUSION] It is steady and feasible to the classification of the different kind species of Fusarium by ITS, but it is difficult to the classification of the different specialized form.
