摘 要:目的:研究哮喘气道重塑大鼠肺组织磷酸化c-Jun氨基末端激酶(P-JNK)的表达,血清/支气管肺泡灌洗液(BALF)白介素1β(IL-1β)的含量,探讨P-JNK在哮喘气道重塑中的作用及IL-1β对JNK活化的可能影响。方法:SD大鼠随机分为对照组(C)、哮喘组(A),复制哮喘气道重塑模型。图像分析技术测定支气管壁厚度(Wat)和平滑肌厚度(Wam),ELISA法测定血清、BALF中IL-1β含量,免疫组化(IHC)、Western blot检测肺内P-JNK蛋白表达,直线相关分析显示Wat、Wam与P-JNK蛋白(平均吸光度mean absorbance,mA表示)的相关性及P-JNK蛋白与血清、BALFIL-1β浓度的相关性。结果:A组Wat、Wam及血清、BALFIL-1β浓度均较C组增加(均P<0.01);A组P-JNK mA及光密度值均较C组增加(均P<0.01);Wat、Wam与P-JNK mA均呈显著正相关(均P<0.01);P-JNK mA与血清、BALFIL-1β浓度均呈显著正相关(均P<0.01)。结论:哮喘气道重塑大鼠肺组织P-JNK、血清、BALFIL-1β表达增强,IL-1β可能促进JNK活化。Objective:To study the expression of phosphorylated c-Jun N-terminal kinase (P-JNK) in lung tissues,the amounts of IL-1β in serum and BALF of asthmatic rats of airway remodeling,whereby to explore the role of P-JNK in asthma airway remodeling and the possible effect of IL-1β on JNK activation.Methods:24 SD rats were randomly divided into the control group(group C) and the asthma group(group A).Asthmatic model of airway remodeling was developed.The total bronchial wall thickness (Wat) and the airway smooth muscle thickness(Wam) were measured by image analysis system.The amounts of IL-1β in serum and BALF were tested by sandwich ELISA.The protein expression of P-JNK was detected by immunohistochemistry.The contents of P-JNK/SAPK in the lung homogenates were detected by western blot.Results:Wat and Wam,the amounts of IL-1β in serum and BALF of group A were all significantly higher than those of group C(P〈0.01,respectively);the P-JNK mA and optical density of group A were both significantly higher than those of group C(P〈0.01,respectively);Wat and Wam were positively correlated with P-JNK mA(P〈0.01,respectively),and P-JNK mA were positively correlated with amounts of IL-1β in serum or BALF(P〈0.01,respectively).Linear correlation analysis showed the correlation between Wat and P-JNK protein(A),Wam and P-JNK protein(A),P-JNK protein(A) and levels of IL-β in serum or BALF.Conclusion:The contents of P-JNK in lung tissues and the levels of IL-1β in serum and BALF are significantly increased in asthmatic rats of airway remodeling,IL-1β probably accelerates activation of JNK kinase.
