摘  要:目的探讨布地奈德(budesonide)雾化吸入对博莱霉素(bleomyein,BLM)致肺纤维化大鼠肺组织羟脯氨酸和Ⅰ、Ⅲ型胶原mRNA表达的影响。方法取健康雌性Wistar大鼠45只,随机分成博莱霉素组(BLM组)、生理盐水(NS组)和布地奈德组(Bud组)各15只,气管内灌注BLM(Bud组、BLM组)和生理盐水(NS组)后0~6d每天雾化1次,Bud组雾化吸入布地奈德,BLM组和NS组雾化吸入生理盐水,各组分别于第7、14、28d处死大鼠各5只。用HE、Masson染色观察肺泡炎症和纤维化改变;肺组织匀浆测定羟脯氨酸含量;逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)测定肺组织Ⅰ、Ⅲ型胶原mRNA表达。结果Bud组第7、14d肺泡炎程度较BLM组减轻(P〈0.05),Bud组第7、14、28d肺纤维化程度均较BLM组减轻(P〈0.05);Bud组第7、14、28d肺组织羟脯氨酸含量较BLM组明显降低(P〈0.05);Bud组第7、14d肺组织Ⅰ、Ⅲ型胶原mRNA表达较BLM组明显降低(P〈0.05)。结论布地奈德雾化吸入可减轻BLM致肺纤维化大鼠肺泡炎症、降低肺组织羟脯氨酸含量和Ⅰ、Ⅲ型胶原mRNA表达,从而减轻纤维化程度。Objective To investigate the effect of inhaled budesonide on the expression of hydroxyproline and type Ⅰ /Ⅲ collagen mRNA in lung tissue of bleomyein-induced rat pulmonary fibrosis. Methods Forty-five adult female Wistar rats were randomly divided into 3 groups:both budesonide(Bud) group and bleomycin(BLM) group were intratraeheally instilled with BLM, and normal solution( NS) group with 0.9% NaCl solution. Both NS group and BLM group were aerosolized with NS,while Bud group with budesonide ev ery day at day 0 - 6 after rats were intratracheally administrated. Each group was again divided into 3 subgroups,which were sacrificed at day 7,14 and 28. Hydroxyproline content was detected;HE and Masson staining were performed to observe the extent of alveolitis and fibrosis;The mRNA levels of type Ⅰ /Ⅲ collagen in the lung tissue were measured by RT-PCR.Results The extent of alveolitis in Bud group at day 7 and 14 were lower than those of BLM group( P 〈 0.05 ). The extent of lung fibrosis and the expressions of hydroxyproline in Bud group at day 7,14 and 28 were significantly lower than those of BLM group ( P 〈 0.05 ) ; The mRNA levels of type Ⅰ/Ⅲ collagen at day 7 and 14 in Bud group were significantly lower than those in BLM group( P 〈 0.05 ). Conclusion Inhaled budesonide could decrease the levels of hydroxyprotine and type Ⅰ /Ⅲ collagen mRNA and ameliorate the formation of pulmonary fibrosis in bleomycin-indueed rat pulmonary fibrosis.
