摘 要:目的研究鹿角灵芝胶囊的抗肿瘤活性及对顺铂所致肾损伤的保护作用。方法建立小鼠Lewis肺癌模型,评价鹿角灵芝胶囊内容物的抗肿瘤和抗氧化作用及对顺铂肾毒性的影响,并用MTT法比较鹿角灵芝胶囊内容物在体外对10种不同类型人肿瘤细胞株的细胞毒活性。结果鹿角灵芝胶囊能明显抑制Lewis荷瘤小鼠的肿瘤生长,并显著逆转顺铂造成的荷瘤小鼠血清Scr与BUN的升高、肾皮质组织MDA含量的升高,以及SOD与GSH-Px活性的降低;鹿角灵芝胶囊对不同类型人肿瘤细胞的细胞毒活性有较大差异,其IC50排序为:HT-29<MCF-7<A549<Hela<Caco-2<SK-OV-3<Eca-109<HL-60<SGC-7901<HePG2。结论鹿角灵芝胶囊具有体内外的抗肿瘤活性,并对顺铂造成的肾损伤有保护作用。OBJECTIVE To investigate the effects of Comu cervi ganoderma lucidum capsule (GLC) on antitumor and kidney dam- age induced by cisplatin in mice. METHODS Mice bearing tumors were randomly divided into normal group, cisplatin (CDDP) group, GLC group, CDDP plus GLC group. The changes of Scr, BUN, MDA, SOD, and GSH - Px of renoeortical tissue were measured and carcinoma inhibition rate were calculated. Inhibition of GLC on 10 different human tumor cell lines were detected by MTr method in vitro. RESULTS In vivo study showed GLC could inhibite the growth of tumors and the contents of Scr and BUN in CDDP group was significantly higher than those in normal group. The content of MDA increased and the activity of SOD and GSH - Px declined in reno- conical tissue. The contents of Scr and BUN in GLC plus CDDP group were significantly lower than that in CDDP group. The content of MDA decleared and the activity of SOD and GSH - Px increased in renocortical tissue. In vitro study, GLC shows different inhibitory effects on different human tumor cell lines. The ICs0 values were as follows : HT -29 〈 MCF -7 〈 3.549 〈 Hela 〈 Caco -2 〈 SK - OV - 3 〈 Eta - 109 〈 HL - 60 〈 SGC - 7901 〈 HepG2. CONCLUSION The results indicated that GLC has the ability of inhibiting tumor growth and could reduce cisplatin induced nephrotoxicity.
