摘  要:目的采用HPLC—FLU测定人血浆中阿司匹林活性代谢物水杨酸的浓度,并研究人体内的药动学。方法采用Zor-baxSB—C18色谱柱(150mm×4.6mm,5μm),流动相为0.02mol·L-1乙酸胺(冰醋酸调pH2.2)-乙腈-四氢呋喃(65:30:5),流速1.0ml·min-1。激发波长为300nm,发射波长为405nm。结果水杨酸0.0288—2.3000μg·ml-1线性良好(r=0.999),最低检测浓度为28.8ng·ml-1,方法回收率92.48%-109.98%,日内、日间RSD分别为3.09%-6.35%、3.08%-7.16%。结论所建方法操作简便、快速、灵敏、准确、干扰少,适用于测定水杨酸血药浓度及阿司匹林药动学研究。OBJECTIVE To establish an HPLC -FLU method for the determination of salicylic acid(SA,the metabolite of aspirin) in human plasma and to apply this method to the pharmacokinetics study of salicylic acid. METHODS The samples were separated on a Zorbax SB - C18 colunm( 150 mm × 4. 6 mm,5 μm)using a mobile phase containing a mixture of 0.02% CH3 COONH3 (adjusted to pH2. 2 with HAc) -acetonitrile -THF(65 : 30 : 5) and a flow rate of 1.0 ml· min-1. Eluents were monitored by fluorescence spectroscopy with excitation wavelength of 300 nm and emission wavelengths of 405 nm, RESULTS The linear range of salicylic acid concentration was0. 0288 -2. 3000 μg·ml-1 (r =0. 999). The limit of detection was 28.8 ng.ml-1 ,recoveries were 92.48% - 109.98%. The variation co- efficients of intra - day(n =5) and inter - day(n =5) were 3.09% -6.35% and 3.08% -7.16% ,respectively. CONCLUSION The method is simple, rapid, sensitive, accurate, and suitable for the study of pharmaeokinetics and bioavailability of aspirin.
