无机化合物CANA对大鼠血管钙化模型的改善作用

摘  要:目的:制备大鼠血管钙化模型并观察枸橼酸盐结构类似的无机化合物CANA对血管钙化的作用。方法:用肌注维生素D3和口服尼古丁诱导大鼠血管钙化模型。4周后,进行血管组织Von Kossa染色,测定大鼠血管钙含量,碱性磷酸酶(ALP)活性、血磷、血钙含量以及心脏系数。结果:模型组大鼠血管VonKossa染色见平滑肌细胞内和细胞外基质有大量银染的深紫色颗粒聚集,主动脉血管钙含量以及ALP活性分别较对照组高66.9%(P<0.001)和462.6%(P<0.001),血磷和血钙含量及心脏系数较对照组比较无明显变化(P>0.05);无机化合物CANA740 mg.kg-1治疗组血管Von Kossa染色的深紫色颗粒有所减少,主动脉血管钙含量及ALP活性分别较模型组降低38.2%(P<0.001)和22.0%(P<0.05)。结论:用维生素D3和尼古丁制备模型4周后,可造成血管组织钙化,但血磷和血钙含量及心脏系数无明显改变。无机化合物CANA对该血管钙化模型有改善作用。Objective : To establish the model of vascular calcification in rats, and observe the effect of the inorganic compound CANA(with similar structure to citrate) on vascular calcification. Methods: Vascular calcification in rats was induced by administration of vitamin D3 plus nicotine. Four weeks late, Von Kossa staining, cal- cium content, alkaline phosphatase aetivity, phosphorus and calcium content in blood, and heart coefficient were assayed. Results : A strong positive staining of dark purple was found among the smooth muscle cells and the extra- cellular matrix in calcified aorta by Von Kossa staining. Calcium content and ALP activity in calcified arteries were increased by 66.9% ( P 〈 0. 001 ) and 462.6% (P 〈 0. 001 ) as compared with the controls. No significant change was tbund in phosphorus and calcium content in blood (P 〉 0.05) , and in the heart eoeffieient(P 〉 0.05). Positive staining of Von Kossa was reduced after treatment of CANA (740 mg. kg^-1 ). The calcium content and ALP activity in calcified arteries treated with CANA (740 mg.kg^-1 ) were dropped by 38.2% (P 〈0. 001 ) and 22.0% (P 〈 0. 05) , respectively. Conclusion: Four weeks after the treatment of vitamin D3 and nicotine, there is only significant calcification in the vascular tissue; while no significant alterations in the phosphorus and calcium contents in blood as well as the heart coefficient. The inorganic compound CANA can improve the vascular calcification in this model.
