摘  要:目的:研究卵泡方在大鼠体内的组织分布规律。方法:卵泡方大鼠灌胃给药6g.kg-1后,应用HPLC测定大黄酸、汉黄芩素及千层纸素A在大鼠肝、心、脾、肺、肾中的含量,色谱柱:Zorbax SB-C18(250mm×4.6mm,5μm),流动相:(A)乙腈-(B)0.5%冰醋酸水溶液(v/v)梯度洗脱;流速为1.0mL.min-1;检测波长为254,276nm;进样量为10μL;柱温30℃。结果:建立了大黄酸、汉黄芩素及千层纸素A在大鼠肝、心、脾、肺、肾中的HPLC含量测定方法。结论:该法成功应用于卵泡方单剂量大鼠灌胃给药后,大黄酸、汉黄芩素及千层纸素A在大鼠体内组织分布的研究。Objective: To investigate the distribution in rat after oral administration of Luan-Pao-Prescription. Methods: Rhein, wogonin and oroxylin A were quantitatively determined by HPLC/DAD method in rat tissues including liver, heart, spleen, lung and kidney after oral administration of Luan-Pao-Prescription. The separation was performed on a Zorbax SB-C18 column (250 mm × 4. 6 mm, 5 μm) with acetonitrile-0. 5% glacial acetic acid aq. (v/v) using linear gradient elution and UV detection at wavelength of 254 and 276nm. The flow rate was 1.0 mL·min^- 1 and the column was kept at 30℃. Results : The HPLC/DAD method for the simultaneous determination of rhein, wogonin and oroxylin A in rat tissues was developed. Conclusion: The method has been successfully applied for the distribution study of three active constituents in rat through oral administration of Luan-Pao-Prescription.
