摘 要:目的:探索ISSR分子标记技术对玉竹的主要栽培品种、野生种及易混淆品黄精进行鉴定的新方法。方法:采用优化的ISSR-PCR体系,筛选出能扩增明显多态性条带的引物对样品进行扩增和凝胶电泳分析,并依遗传距离构建分子聚类图。结果:10个引物共扩增出125个DNA片段,其中116个片段呈现多态性,多态带百分率为92.9%。玉竹品种间存在较大的遗传多样性,栽培品种与野生种及易混淆品黄精易于区分。结论:本实验所建立的ISSR分子标记方法可用于玉竹的品种鉴定和良种选育研究。Objective: To establish a molecular method by ISSR for the identification of the Polygonatum odorarum including cultivated strain, wild strain and its adulterant, notheds: Primers with polymorphic and informative patterns were selected to amplify for these samples. The Nei' s genetic distance is obtained based on the result, then UPGMA cluster analysis is using by the Nei' s genetic distance. Results: 125 bands were obtained by 10 ISSR primers. Of which, the polymorphic bands are 116, the percentage is 92. 9%. There was a rich genetic diversity among the Polygonatum odoratum. The cultivated, wild and Polygonatum odoratum adulterant are identified easily. Conclusion: The ISSR could be applied to identify and select good strain among Polygonatum odoratum.
