摘 要:【目的】分离、纯化和鉴定万隆霉素抗菌作用的主要活性成分。【方法】以体外抗枯草芽孢杆菌为活性跟踪指标,通过硅胶柱层析和高效液相色谱等方法,从万隆霉素产生菌GAAS 2507发酵物中分离得到抗菌主要活性成分,采用电喷雾质谱(ESI-MS)、核磁共振谱(1HNMR、13CNMR、DEPT、DQFCOSY、HSQC、HMBC、NOESY)波谱法对其进行结构解析。【结果】波谱结构解析表明抗菌活性成分为Quinomycin C,其对细菌性条斑病菌(MIC值为1.563μg·ml-1)、黄瓜疫病菌(EC50值为1.256μg·ml-1)和节瓜枯萎病菌(EC50值为20.570μg·ml-1)具有明显的抗菌活性。【结论】Quinomycin C是万隆霉素主要活性成分之一。[Objective] To separate, purify and identify the antimicrobial component of Wanlongmycin. [Method] Using Bacillus subtilis as an indicator strain, an autimicrobial component was obtained from culture broth of Wanlongmycin producing strain GAASS 2507 by silica gel chromatography and HPLC, and its structure was identified by means of spectroscopic anaysis such as MS, 1DNMR, 2DNMR. [Result] The active component was indicated to be Quinomycin C by its spectral data and its MIC values against Xanthomonas campestris pv. Oryzicola was 1.563μg·ml^-1, EC50 values against Phytophthora melonis and Fusarium oxysporum f.s.p, cucumerinum owen were 1.256μg·ml^-1 and 20.570μg·ml^-1, respectively. [Conclusion] It is concluded that Quinomycin C is one of the main antimicrobial components and its inhibition towards phytopathogenic fungi and bacteria is reported for the first time.
