摘 要:以体外培养的RPMVECs(rat pul monary microvascular endothelial cells,RPMVECs)为模型,采用双夹心ELISA法和MTT法,研究了绿原酸、连翘酯苷、黄芩苷、黄芩素、汉黄芩素诱导RPMVECs表达IFN-γ(inferteron-γ)的影响以及对PMVECs的毒性。ELISA检测结果表明,加入质量浓度为5,10,20,50,100μg/mL的5种中药成分刺激细胞16 h后,在培养细胞上清液内均能检测到IFN-γ,其中绿原酸、黄芩素组IFN-γ的表达量随药物浓度增大而减少;连翘酯苷组IFN-γ的表达量均较多;黄芩苷、汉黄芩素组均在10μg/mL时IFN-γ的表达量达到峰值,随后IFN-γ的表达量随浓度增大而减少。MTT检测结果显示,各浓度的黄芩苷、黄芩素对PMVECs均有一定程度的抑制作用,而连翘酯苷对PMVECs均有增殖作用,绿原酸、汉黄芩素的细胞毒性则随药物浓度而发生变化。The model was based on cultured rat pulmonary microvascular endothelial cells in vitro. With the methods of double antibodies sandwich ELISA and MTT, the influences of chlorogenic acid, forsythiaside, baicalin,daicalein and wogonin on inducing PMVECs to express inferteron-γ and their cytotoxicity were studied. The result of ELISA indicated that the mass concentration of five constituents were 5,10,20,50 and 100 μg/mL,at 16 hours after added,IFN-γ in the cell supernate fluids had all been detected. In groups of chlorogenic acid and daicalein,the expression of IFN-γ decreased as drug level rised while it was all relatively higher in group forsythiaside. In groups of baicalin and wogonin,IFN-γ level reached peak at 10 μg/ mL, then reduced. The MTT results showed that baicalin and daicalein had some inhibitory action at different concentrations PMVECs while the action of forsythiaside was the opposite. The cytotoxicity of chlorogenic acid and wogonin varied at different drug concentrations.
