摘  要:肌肉生长抑制素是控制骨骼肌生长发育的重要细胞因子,采用PCR-SSCP和测序方法分析了大黄鱼肌肉生长抑制素基因外显子I的单核苷酸多态性,结果发现,外显子I有7个多态性位点,分别是270(G→A)、196(C→T)、349(A→T)、51(T→C)、352(G→A)、348(C→T)和264(G→T),但只有349位点和352位点的碱基突变造成错义突变,氨基酸变化分别为T→S和E→K,其他位点均属同义突变。存在7种基因型,它们的频率分别为0.2708、0.0833、0.3333、0.0208、0.1667、0.1047和0.0208,该基因位点的杂合度为0.709。统计分析表明,外显子I无论是基因型水平上的多态性还是氨基酸水平上的突变均与体长和体质量呈显著性相关(P<0.05),但与肌肉脂肪含量无显著相关(P>0.05)。Myostafin (MSTN) as an important cytokine controls growth and development of skeletal muscle in animals. Seven single nucleotide polymorphisms (SNPs) were totally identified in the exon Ⅰ of MSTN gene from large yellow croaker Pseudosciaena crocea by PCR-SSCP and sequencing. Seven polymorphic sites were located at 270 ( G→A ), 196 ( C→T), 349 ( A→T), 51 ( T→C ), 352 ( G→A ), 348 (C→T) and 264 (G→T). Base mutations at sites 349 and 352 resulted in amino acid substitutions of T→S and E→K, respectively. However, base mutations at other sites did not cause amino acid substitution. There were seven genotypes in the examined population, whose frequency was 0.2708, 0.0833, 0.3333, 0. 0208, 0. 1667, 0. 1047 and 0.0208, respectively. The heterozygosity of MSTN locus was 0.709. Statistics analysis showed that the differences in both genotypes and amino acid sequences had a significant effect on the body length and weight of the fish, but had no significant effect on the total lipid content in skeletal muscles.
