摘 要:目的:探讨糖基化终产物在糖尿病牙周病形成中的作用。方法:原代培养牙周膜成纤维细胞,将糖基化终产物修饰的人血清白蛋白及未修饰人血清白蛋白与牙周膜成纤维细胞在体外共同培养,MTT法测定细胞增殖,用碱性磷酸酶测定法测定ALP。结果:糖基化终产物修饰的人血清白蛋白能以浓度依赖的方式抑制牙周膜细胞的增殖(p<0.05),而未修饰人血清白蛋白可使细胞增殖,但两种白蛋白均不诱导ALP的表达。结论:糖基化终产物可通过抑制牙周膜成纤维细胞的增殖,降低糖尿病病人牙周组织对损伤的修复能力,导致牙周病。Objective:periodontitis disease is one of the most important complication in patients with diabetes mellitus. Advanced glycation end products (AGE) modified proteins are found in plasma of diabetes mellitus patients. The study was conducted to elucidate the infection of AGE on proliferation of human periodontal ligament fibroblasts.Method:Human periodontal ligament fibroblasts were coincubated in vitro with native human serum albumin(HSA),HSA modified with advanced glycation end products(AGE-HSA). The proliferation of human periodontal ligament fibroblasts was determined by MTT and ALP of human periodontal ligament fibroblasts was determined.Result:AGE-HSA inhibited the proliferation of human periodontal ligament fibroblasts in a dose-dependent manner(P〈0.05).HSA increased the proliferation of human periodontal ligament fibroblasts. AGE-HSA had no effect on the expression of ALP.Conclusion:AGE inhibited the proliferation of human periodontal ligament fibroblasts and this can delay the periodontal tissue repaired.
