摘 要:目的了解精液中生精细胞的状况和生精细胞凋亡率与不育症的关系,并作为评估睾丸功能的指标。方法293例精液标本用瑞-姬染色,油镜下进行生精细胞形态学及凋亡率检出与分析。并采用DNA缺口原位末端标记技术(TUNEL)染色进行对照,确认瑞-姬染色对生精细胞凋亡鉴别的可行性。结果在293例精液中,有生精细胞者273例占93.1%;无生精细胞者20例占6.9%。生精细胞凋亡率随精子数量减少而增加,以无精子症者最高,比生育组高5.8倍(P<0.001)。显示瑞-姬染色的可行性与优越性。结论睾丸生精障碍与生精细胞过度凋亡密切相关,可在精液中检出脱落生精细胞,根据精液生精细胞凋亡率可作为评估睾丸功能的指标之一。Objective: To understand the state of spermatogenic cells in semen and the relationship between apoptosis rate and infertility. Methods: Semen samples were collected from 293 infertility men and dyed with the Wright' s-Giemsa' s Stain. The morphological characteristics and apoptosis rate of the spermatogenic cells were examined. The TUNEL assay was used to confirm the feasibility of Wright's-Giemsa's Stain for the identification of spermatogenic cell apoptosis. Results: The spermatogenic cells were found in 273 of 293 (93.1%) semen samples. The spermatogenic cell apoptosis rate increased with the decreased sperm quantity. The highest apoptosis rate was found in those samples without sperm, which was 5. 8 times as much as that of fertile group (P〈0. 001). Wright's-Giemsa's Stain was a convenient and feasible way to detect the apoptosis of spermatogenic cells in routine lab test. Conclusions: The spermatogenesis impairment is closely related with the over-apoptosis of spermatogenic cells. The shed spermatogenic cells can be detected in semen and their apoptosis rate may be used as an indicator of testicular function.
