摘  要:目的研究马兜铃酸(AA)作用下大鼠肾小管上皮细胞尾加压素Ⅱ(UⅡ)含量的变化。方法采用机械分离和酶消化法获取大鼠肾小管上皮细胞并进行培养、鉴定;用RT-PCR和Western blot法分别测定不同浓度(0ng/ml、5ng/ml、10ng/ml、20ng/ml)AA作用下大鼠肾小管上皮细胞中Ⅶ基因和蛋白表达情况。结果培养的细胞经Cytokeratin-18免疫细胞化学染色确定为肾小管上皮细胞。AA以剂量依赖的方式促进肾小管上皮细胞UIImRNA和蛋白表达,5ng/ml AA作用下肾小管上皮细胞UⅡ mRNA表达即显著增强(P<0.01),其蛋白表达在10ng/ml AA作用下明显增强(P<0.05)。结论AA作用下大鼠肾小管上皮细胞UⅡ含量明显增加,推测UⅡ在AA所致大鼠肾小管上皮细胞损伤中可能有一定作用。Objective To explore the effect of variety concentrations of aristolochic acid (AA) on urotensin Ⅱ(UⅡ) in rat renal tubular epithelial cells. Methods The rats renal tubular epithelial cells were acquired with mechanical dissociation and enzyme digestion, which be cultured and identified sequently. The effect of different concentrations of AA (0 ng/ml,5 ng/ml, 10 ng/ml,20 ng/ml) on expression of UⅡ gene and protein in rat renal tubular epithelial cells was observed, which through the methods of RT-PCR and Western blot, respectively. Results Renal tubular epithelial cells was stained by cytokeratin-18 immunocytochemical. It showed that dose dependent effect of AA on expression of UⅡ mRNA and protein. With 5 ng/ml AA, the expression of UⅡ mRNA in rat renal tubular epithelial cells demonstrated remarkably ( P 〈 0.01 ). With 10 ng/ml AA, the expression of UⅡ protein increased significantly ( P 〈 0.05). Conclusion It proved that the UⅡ in rat renal tubular epithelial cells increased obviously with the effect of AA. It suggested that UⅡ probably have some effects on the rat renal tubular epithelial damaged cells induced by AA.
