摘 要:应用PCR方法从疑似断奶仔猪多系统衰竭综合征(PMWS)的死亡仔猪组织病料中扩增出PCV-2全基因组(1767bp),将此基因片段克隆入pMD20-T载体,筛选获得重组质粒pMD-PCV2并对其进行序列测定,然后对全基因组进行同源性和遗传进化分析。结果表明,PCV-2分离毒株与参考株的全基因组同源性介于94%~98.4%之间,ORF1的同源性介于97.0%~98.8%,ORF2的同源性略低,介于91.9%~98.4%之间。该毒株与荷兰株、广西株和上海株等在一个进化分支上。本研究有助于监测PCV-2的疫源和进化关系,为进一步深入研究病毒基因功能奠定一定基础。The full-length nucleotide sequence of PCV-2 was amplified by PCR from the tissues of clinical pigs with postweaning multisystemie wasting syndrome. The fragment amplified was cloned into pMD 20- T vector and the recombinant plasmid was sequenced and determined. Then, the homology and evolutional relationship of PCV-2 genome and reference strain sequences were analyzed. The results of homology analysis showed that genome of Hebei isolate shared 94.0 %-98.4 % homology with reference sequence and the ORF1 exhibited 97. 0%-98. 8% sequence similarity, but the ORF2 showed lower homology, 91. 9%- 98.4%. Phylogenetic analysis revealed that the Hebei strain is clustered in the PCV-2. The study would play a foundation to understand the genic function of the virus.
