摘  要:以北京小黑豆未成熟子叶为外植体,针对影响体细胞胚发生的几个关键因素进行研究。结果表明:pH等于7,外植体大小为2—3mm和暗培养条件最适合体细胞胚的发生。利用优化后的再生系统诱导了北京小黑豆(再生性强)和Keburi(再生性极低)杂交所衍生的150个F5:6重组自交系,体细胞胚发生频率明显呈现间断分布,由此初步推断大豆体细胞发生频率由少数几个基因控制。由体细胞胚发生效率分布偏向Keburi,可见Keburi对体细胞胚发生效率影响要比北京小黑豆大。With Peking immature cotyledon as explants, key factors that affecting somatic embryogenesis were evaluated. The results showed that pH = 7.0, explant size of 2- 3 mm and dark culture conditions were beneficial for the occurrence of somatic embryogenesis, which was proved efficient for somatic embryogenesis. One hundred and fifty F5:6 recombinant inbred lines derived from Peking × Keburi was cultured with inducing medium for embryogenesis. The results showed that frequency distribution was discrete, suggesting that embryogenesis was controlled by few genes. Meanwhile, the impact of Keburi on embryogenesis efficiency was higher that that of Peking.
