摘  要:目的观察联合应用COX-2抑制剂塞莱西布和免疫佐剂CpG-ODN对树突状细胞(DC)功能的影响,研究其生物效应。方法分离和培养人脐血来源树突状细胞,按分组进行预处理和诱导成熟,光镜及电镜观察DC形态,流式细胞仪检测表面标志CD1a、CD83、HLA-DR的表达情况以及进行细胞因子IL-10I、L-12水平测定(ELISA)。结果按预定方法诱导成熟的DC,其形态学无明显异常,流式细胞检测证实了其正常成熟并具有生物活性,细胞因子的测定表明实验组IL-12含量明显增高,IL-10含量明显降低,Th1/Th2应答平衡出现明显的免疫漂移现象。结论联合应用COX-2抑制剂和CpG-ODN可明显增强DC的生物学活性,为建立基于DC的肿瘤生物治疗打下了良好的基础。Objective To observe the contribution on function of dendritic cell by using COX-2 Inhibitor, eelecoxib, and CpG- ODN,and study the biological effect. Methods DC from humen cord blood was separated,cultivated,preconditioned and induced to maturity. The appearance of DC was observed by light microscope and electron microscope. Expression of surface markers including CD1a,CD83 and HLA DR were detected by flowcytometry. Then the level of cytokine IL10 and IL-12 were detected by ELISA. Results The appearance of DC was normal. It was confirmed that DC has more powerful bioactivity and it could lead to immuno logical drift obviously. Conclusion The biological effect of DC has been enhanced by combining celecoxib and CpG-ODN. Better prospect should be anticipated in tumor theropy based DC.
