Changes of serum and BALF IL-6 level and their effect on the pathogenesis of acute lung injury

摘  要:采用白细胞介素-6(Interleukin6,IL-6)依赖细胞株7TD1及MTT比色法,动态观察了正常及内毒素注射后不同时间大鼠血清和支气管肺泡灌洗液(bronchoalveolarlavagefluid,BALF)中IL-6含量的变化;同时,还对各组大鼠的肺体指数和BALF中蛋白质含量进行了测定。结果显示,大鼠注射内毒素后1、3、6、12h,其血清和BALF中IL-6含量急剧升高(P<0.01),注射后24h降为正常(P>0.05);此外,其肺体指数和BALF中蛋白质含量亦明显高于对照组,尤以3h和6h显著(P<0.01);且内毒素致伤后大鼠血清和BALF中IL-6含量的升高与BALF中蛋白质含量的升高呈显著正相关。推测IL-6可能参与了肺水肿的形成,其机制有待进一步探讨.Changes of IL-6 contents in serum and bronchoalveolar lavage fluid (BALF) in rats of normal and after endotoxin infusion were measured using the IL-6 dependent cell line 7TD1 and MTT colorimetric assay, and the lung-body index and protein contents in rat's BALF were also measured. The results showed that the serum and BALF IL-6 contents were sighficantly higher (P<0.01) at 1, 3,6, 12h after endotoxin infusion and returned to the baseline at 24h after endotoxin. Moreover, the lung-body index and protein content in Bas were also increased sharply after endotoxin infusion, especially at 3,6h after infusion. There were a positive relationship betWeen the IL-6 content in plasma or BALF and the protein content in BALF after endotokin infusion. The findings indicated that elevated serum and BALF IL-6 level is important in the pathogenesis of acute lung injury.
