摘 要:目的比较3种不同树脂纯化细胞色素C(CytC)的效果。方法以健康猪的新鲜心肌为材料,采用酸化水提取,等电点沉淀杂蛋白,人造沸石吸附,硫酸铵盐析,三氯乙酸沉淀,透析过滤后得到CytC粗品,再采用国产D-152、724弱酸性阳离子交换树脂、以及进口AmberliteGC-50等3种不同的离子交换树脂对CytC进行纯化,纯化后的CytC加入少量联二亚硫酸钠,通过分光光度法测定其浓度,并计算收率。结果采用国产D-152树脂的收率为85.09%,724弱酸性阳离子交换树脂的收率为82.54%,AmberliteGC-50的收率为91.97%。结论3种树脂中Amber-liteGC-50纯化CytC的效果最好。Purpose To compare the effects of three different cation exchange resins for purifying Cytochrome C. Methods Cytochrome C was extracted from health pig heart muscle by acid liquid water, isoelectric point precipitation, Permuter absorption, ammonium sulfate salting out, TCA precipitating, dialysis and filtering, and the Cytochrome C was purified by D-152, 724 and Amberlite GC-50 cation exchange resin. Then the Cytochrome C was scaned in 520 nm, the recovery calculated. Results The recovery of D-152 was 85. 09%, 724 was 82.54%, and Amberlite GC-50 was 91.97%. Conclusion Amberlite GC-50 is the best for purifying Cytochrome C.
