复方蒲黄颗粒对大鼠脑缺血再灌注损伤的保护作用

摘　　要：目的研究临床有效的中药复方蒲黄颗粒对大鼠脑缺血再灌注损伤的保护作用。方法实验采用90只清洁级SD大鼠,随机分为6组(每组15只):假手术组(sham组),模型组(I/R组),复方蒲黄颗粒大剂量组(PHH组),复方蒲黄颗粒中剂量组(PHM组),复方蒲黄颗粒小剂量组(PHL组),脑血栓片组(NXS组)。采用右侧颈总动脉结扎手术,建立大鼠脑缺血再灌注损伤模型,分别在术后6h、24h、72h通过神经功能评分观察神经行为学改变,采取TTC染色计算脑梗死体积。结果①与sham组相比,其他各组在术后6h评分均较高,并随时间推移评分逐渐下降;与I/R组比较,PHL组、NXS组评分无显著改善(P>0.05),但在PHH组、PHM组评分则有显著改善(P<0.05)。②I/R组梗死体积为21.55%;PHL组、PHM组、PHH组、NXS组梗死体积在12.87%～15.01%波动,与I/R组相比较,梗死体积显著缩小(P<0.05)。结论复方蒲黄颗粒对大鼠脑缺血再灌注损伤具有良好的保护作用,以大、中剂量作用最为显著。Objective To study the protective effects of ＂Fufang Puhuang Granule＂ on cerebral ischemia-reperfusion injury in rats. Methods Ninety Sprague-Dawley rats were randomized into six groups（15 rats in each group）： sham group, cerebral ischemia-reperfusion group（I-R group）, high-dose ＂Fufang Puhuang Granule＂ group （HPH group）, medium-dose ＂Fufang Puhuang Granule＂ group （MPH group）, low-dose ＂Fufang Puhuang Granule＂ group （LPH group）, Naoxueshuan group （NXS group）. The model of cerebral ischemia-reperfusion injury was established by fight common carotid artery occulusion; neurological grade evaluation was used to examine the behavioral recovery at 6 hours （6 h）, 24 hours （24 h） and 72 hours （72 h） after reperfusion, and the volume of cerebral infraction was determined with TTC staining. Results ①Compared with sham group, the scores of neurological grade 6 hours after isehemia-reperfusion in other groups were increased, but they decreased with time; compared with I-R group, the scores were not increased in LPH group and NXS group （P 〉 0.05）, but they were increased in HPH and MPH groups （P 〈 0.05）. ②The volume of cerebral infraction in I-R group was 21.55%, but it was between 12.87%-15.01% in LPH group, MPH group, HPH group and NXS group, all lower than I-R group（P 〈 0.05）. Conclusion ＂Fufang Puhuang Granule＂ is quite effective in preventing cerebral ischemia-reperfusion injury, in particular in high- and medium-dosage.
