摘 要:对来自东海-黄海海区10个站位以及青岛沿岸4个站位共14个站位的浒苔样品依据形态学特征,归类分离获得70个样品,对其ITS+5.8S rDNA区进行PCR扩增、测序和分析,并与NCBI数据库中相应序列进行了比对。结果表明,70个样品具有完全相同的ITS和5.8S rDNA序列,说明青岛沿岸漂浮的大量浒苔与东海-黄海海区的浒苔为同一个来源。聚类分析显示,浒苔ITS区的变异程度与地理间隔距离呈正相关,我国海域浒苔与日本海域浒苔聚类一起,与英国海域浒苔相聚较远,与美国海域浒苔距离最远。Enteromorpha prolifera samples were collected from 10 sites in the East Sea-Yellow Sea area and four sites in Qingdao. The samples were sorted into five categories based on the morphological characters. ITS+5.8S rDNA region of a total of 70 samples were amplified and sequenced. The ITS + 5.8S rDNA sequences obtained were compared to those downloaded from NCBI genBank. The results showed that the ITS +5.8S rDNA sequences of the 70 samples were identical, which fell into the same cluster of the E. prolifera isolates from Japan. The divergence of ITS region of the samples was positively correlated to their geographical distance. E. prolifera isolates studied in this experiment were most close to those from Japan, next to those from the United Kingdom and then those from the United States. It was concluded that the mass floating E. prolifera was of the same origin with those in the East and Yellow Sea.
