摘 要:[目的]探讨焦化厂工人ERCC8基因多态性与外周血淋巴细胞DNA损伤的关系。[方法]选择某焦化厂多环芳烃(PAHs)接触者207名焦炉作业工人和115名无PAHs接触及其他毒物暴露者,采集外周静脉血,分离淋巴细胞.使用单细胞凝胶电泳实验观察DNA损伤情况;采用TaqMan-MGB探针检测ERCC8基因分型:运用PHASE2.0.2遗传分析软件计算单倍型。[结果]接触组外周血淋巴细胞的Olive尾矩(Olive Tail Moment,OTM)高于对照组(经对数转换后分别为1.25±1.17和0.53±0.98),差异有极显著性(P<0.01)。在接触组中,ERCC8基因Rs3117位点CT+CC和TT基因型携带者外周血淋巴细胞的OTM分别为1.57±1.13和1.15±1.67,差异有统计学意义(P<0.05);启动区Rs158920位点CT+TT和CC基因型携带者外周血淋巴细胞的OTM分别为1.14±1.21和1.24±1.17,差异无统计学意义(P>0.05)。对照组中相应位点的OTM分别为0.54±0.93和0.56±1.00以及0.52±0.86和0.54±1.02,差异均无统计学意义(P>0.05)。在接触组中,两个位点组成的单倍型对中CT/CT单倍型对和其他单倍型对携带者外周血淋巴细胞的OTM分别为1.20±1.17和1.36±1.17;对照组分别为0.54±1.02和0.51±0.91,两组差异均无统计学意义。[结论]接触组外周血淋巴细胞ERCC8基因Rs3117位点的不同基因型和DNA损伤的严重程度有关联。[ Objective ] To study the relationship between ERCC8 gene polymorphisms and DNA damage in lymphocytes of coke oven workers. [ Methods ] 207 coke oven workers and 115 employees who had no exposure history to PAHs and other toxicants were selected as controls. Lymphocytes were separated from their peripheral venous blood. Single cell gel electrophoresis was used to evaluate DNA damage, and TaqMan-MGB probes were used to analyze ERCC8 gene polymorphisms. Using PHASE 2.0.2 software to calculate haplotype. [ Results ] The lymphocytes olive tail moment ( OTM ) of coke oven workers was significantly higher than that of controls( 1.25 ± 1.17 vs 0.53 ± 0.98, P 〈 0.05 ). In coke oven workers group, the OTM of those with CT+CC genotype at ERCC8 gone Rs3117 was1.57 ± 1.13, which was significantly higher than that of those with TT genotype( 1.15 ± 1.67, P 〈 0.05 ). The OTM had no significantly difference between CT+TT and GG genotype at Rs158920 site. In the control group, The OTM had no significantly difference at Rs3117 and Rs158920sites. The OTM of both coke oven workers and controls had no significantly difference between TC/TC Haplotype and other Haplotypes. [ Conclusion ] The gene polymorphism at ERCC8 gone Rs3117 site had association with severe DNA damage.
