摘 要:[目的]研究软骨藻酸诱导PCl2细胞凋亡及对活性氧的影响。[方法]流式细胞仪法测定0、0.01、0.1、1μmol/L软骨藻酸作用PCl2细胞24h后,对细胞凋亡、细胞周期和活性氧生成的影响。[结果]软骨藻酸可诱导PCl2细胞凋亡的发生,与对照组(凋亡率为0.250±0.053)相比,所有染毒浓度组PCl2细胞凋亡率均上升且差异有统计学意义;并将PCl2细胞阻滞于细胞周期中的G2/M期,0.1、1μmol/L组中处于G2/M期的PCl2细胞数量与对照组相比,差异具有统计学意义;所有浓度组均可使PCl2细胞的活性氧分子(reactive oxygen species,ROS)平均荧光强度增加(0.01μmol/L组为187.140±3.629,0.1μmol/L组为206.023±5.277,1μmol/L组为348.915±3.343),与对照组(163.660±4.059)相比,差异均具有统计学意义。[结论]软骨藻酸能诱导PCl2细胞凋亡的发生,将PCl2细胞阻滞于细胞周期中的G2/M期,并使得细胞内的ROS产生增加。[ Objective ] To study the apoptosis of PC12 cells induced by domoic acid and the effect of this acid on the cell cycle and reactive oxygen species of the same cell. [ Methods ] The apoptotie PC12 cells, treated with 0, 0.01, 0.1, 1 μmol/L domoic acid separately for 24 h, and the cell cycle as well as reactive oxygen species were measured by the Flow Cytometrie Analysis. [ Results ] Domoic acid could increase the rate of apoptosis of PC 12 cells in all dose groups, compared with the control group in which the rate of apoptosis was only 0.250 ± 0.053. The cell cycle was blocked markedly at G2/M phase after exposing PC12 cells to domoic acid for 24h specifically in the 0.1, 1 ±mol/L groups. Domoie acid also had an effect on ROS in PC12 cells, and all the dosage groups exhibited significant elevated ROS level than in the control group( the fluorescence intensity was 163.660 ± 4.059 in the control group, whereas was 187.140 ± 3.629 in 0.01 μmol/L group, 206.023 ± 5.277 in 0.1 μmol/L group, and 348.915 ± 3.343 in 1 μmol/L group ). [ Conclusion ] Domoie acid could induce apoptosis of PC12 cells, block the cell cycle at G2/M phase and elevate the ROS level in PC12 cells.
