摘  要:目的:探讨内源性CO和NO在大鼠百草枯(PQ)中毒性急性肺损伤中的作用。方法:肺组织常规HE染色和取血测定血浆丙二醛(MDA)含量来评价肺组织损伤情况;检测血浆中肿瘤坏死因子(TNF-α)水平。检测肺组织中CO和NO含量和诱导型一氧化氮合酶(iNOS)活性。Western Blot方法测定HO-1和iNOS表达的变化。结果:PQ中毒后大鼠肺组织损伤明显加重,血浆MDA含量及TNF-α水平明显升高,应用正铁血红素(Hm)后血中CO含量明显升高,iNOS活性和NO水平则降低。结论:大鼠百草枯中毒性急性肺损伤时内源性CO和NO产生增加,在应用CO的供体氯血红素后CO升高更为明显,而NO产生降低,在一定程度上减轻了百草枯所致的大鼠急性肺损伤。Objeetive..To investigate the effect of endogenous CO and NO in rats with paraquat induced lung in jury. Methods: The injured lung tissue was studied by HE staining and plasma MDA concentration was detected to evaluate the severity of lung injury. Level of TNF-α was detected. The content of CO, NO and the activity of iNOS in the lung tissue was determined. The expression of HO-1 and iNOS protein was detected by Western Blot. Results: The degree of lung injury was serious obviously after paraquat poisoning. The content of MDA and level of TNF-α elevated significantly. After treatment with heroin, the content of CO increased, but the activity of iNOS and the content of NO decreased. Conclusion: Endogenous CO and NO in lung tissue were increased in rats with paraquat induced lung injury. The level of CO was increased and NO was decreased after treated with hemin, and it might lessened the parauqat induced acute lung injury of rats.
