摘 要:为获得高絮凝活性菌株,采用低能氮离子诱变方法,对生物絮凝剂产生菌FJ-7进行诱变选育,筛选得到一株絮凝活性高、遗传稳定性良好的突变株 NIM-192。发酵产絮凝剂曲线表明,其菌体生长速度稍慢于原始菌株,但絮凝活性一直高于原始菌株,絮凝率比原始菌株提高了34.26%。进一步研究表明,蔗糖、葡萄糖、半乳糖是NIM.192产生絮凝剂的适宜碳源,酵母膏:尿素为1:1的混合氮源为最佳氮源,培养基起始 pH 为7~9时,发酵液的絮凝效果最好。NIM-192, a bioflocculant-produeing strain with good genetic stability and high flocculating activity, was screened out through implanting N^+ into FJ-7, its original strain. Its curve of bioflocculant-producing by fermentation showed the growing speed of NIM-192 was slower, but the flocculating activity of bioflocculant was higher than the original strain, and the flocculating rate of NIM-192 increased by 34.26 %. Further studies showed that sucrose, glucose, galactose were the suitable carbon sources, the optimum nitrogen source was the mixture of yeast extract and urea with the proportion of 1 : 1, and the best flocculation effect was obtained when the initial solution pH was adjusted to 7 - 9.
