摘  要:目的观察不同冷冻方法对胎儿卵巢组织早期卵泡的影响。方法分别采用程序冷冻法(PFG)、快速冷冻法(RFG)、玻璃化冷冻法(VG)冻存胎儿卵巢组织,解冻后进行光镜、电镜观察。结果光、电镜组织形态学观察发现冻后各组卵泡结构均存在一定程度的冷冻损伤。冷冻后的胎儿卵巢内形态正常的卵泡数明显下降,在2 mm×2 mm×1 mm的卵巢皮质内,形态正常的卵泡数在PFG、RFG、VG及新鲜对照组(FCG)中分别为(992.3±618.2)个、(542.8±202.5)个、(1604.8±964.0)个和(1888.2±645.3)个。RFG卵泡数显著低于FEG与VG(P<0.05),但与PFG间差异无统计学意义(P>0.05)。结论三种冷冻方法均可有效冻存胎儿卵巢组织,其中玻璃化法对卵巢内早期卵泡的损伤最小,单位体积内保存的卵泡数最多。Objective To distinguish the effects of three different freezing methods on morphology of fetal ovary tissue. Methods The fetal ovary tissue were respectively preserved by program freezing, rapid freezing, and vitrification. After being thawed, fetal ovary tissues in each of these groups were assessed by using beth light and electron microscopy. Results The post - thawing follicular structure was found, to some extend, injured when being observed histomorphologieally by me, arts of light and electron microscopy. The number of morphologically normal follicles was found significantly reduced in post - thawing fetal ovaries. In an unit volume of 2 mm × 2 mm×1 mm dissections,the number of morphologically normal follicles is 992.3 ±618.2,542.8 ±202.5,1604.8 ±964.0 and 1888.2 ±645.3 in each freezing group and the fresh transplanting group. The number of follicles in RFG was notably lesser than that in FCG or in VG( P 〈 0.05), but no significant difference was found between RFG and PFG( P 〉 0.05). Conclusion The three different freezing methods can all effectively preserve fetal ovary tissue.Among the three freezing methods, vitrification has the least frozen injury of early follicles, and it preserve the most follicles per unit volume.
