贯穿缝扎法制作兔耳缺血淤血模型的研究

摘　　要：目的建立一种制作简单、便于管理、可靠的缺血淤血动物模型。方法新西兰大白兔6只,贯穿兔耳背腹侧皮肤和软骨,间断缝扎除尾侧血管外所有动、静脉,缝扎处暴露并外涂碘伏每天3次。分别在缝扎后第1、3、7、14、21天对比观察兔耳皮温、色泽、水肿和缝扎处感染情况,同时行静脉血气分析。结果缝扎后兔耳马上出现明显紫黑色,14天左右变浅,至21天时兔耳皮肤仍为淡紫黑色。缝扎后第1天兔耳开始出现水肿且逐渐加重至7天最为明显,此后逐渐减轻,21天时已无明显水肿。皮温、氧分压、二氧化碳分压在各观察点均较缝扎前有明显下降(P<0.01)。结论贯穿缝扎方法制作的兔耳缺淤血模型可靠、制作和管理简单,可作为缺血淤血所致创面的修复研究模型。Objective To construct an animal model for ischemia and blood stasis by ligation.Methods Six New Zealand white rabbits were selected to pass the full-layer skin and cartilage of rabbit ears and to ligate all blood vessels except for those from caudate. The location of suture was exposed and sterilized by iodophors three times a day. The changes of temperature and color, as well as dropsy and infection of suture part were observed, and the vein blood gas analysis for the ears was performed on day 1, 3, 7, 14, 21 after ligation.Results The color turned to deep violet-black immediately after stitch and lighter 14 days later, still somewhat violet-black till 21 days. Dropsy appeared on the first day and was very obvious on the seventh day after ligation, thereafter the degree of dropsy gradually decreased and dropsy was not noticeable on 21st day after ligation. The parameters we detected such as temperature, oxygen partial pressure and carbon dioxide partial pressure of vein blood were lower after ligation than before ligation （P〈0.01）.Conclusion The model we established is reliable and convenient to produce, which can be used in the study on the wound repair resulting from ischemia and blood stasis.
