摘 要:目的观察低氧对人肺动脉平滑肌细胞(pulmonary arterial smooth muscle cell,PASMC)细胞表型的影响以及不同细胞表型下蛋白激酶G Ia(protein kinase GIa,PKGIa)mRNA及其蛋白表达水平变化,探讨PKGIa信号途径与低氧PASMCs表型转换中的可能调控作用。方法组织块法培养人PASMC。采用RT-PCR和免疫细胞化学法检测常氧组(N组)、低氧12、24h组PASMCs内平滑肌α肌动蛋白(smooth muscle α actin,SM-α-actin)mRNA及蛋白的表达水平变化;同时采用RT-PCR及Western blot检测PKGIa基因mRNA以及相应的蛋白的表达水平。结果各组均检测出SM-α-actin、PKGIa的mRNA以及蛋白表达的变化。低氧刺激下,PASMCs内SM-α-actin的mRNA及蛋白表达水平明显降低;同时PKGIa的mRNA以及蛋白表达表达逐渐降低。结论PKGIa可能在低氧致人PASMCs表型改变中有重要的调控作用。Objective To observe the changes of cell phenotype and PKGIa expression in pulmonary arterial smooth muscle cells (PASMCs) induced by hypoxia, and to investigate the role of PKGIa signaling pathway in hypoxia-induced pulmonary hypertension (HPH).Methods PASMCs were isolated and cultured by tissue-sticking methods. Semi-quantitative reverse transcription and polymerase chain reaction (RT-PCR) were used to examine the PKGIa and smooth muscle α actin (SM-α-actin) mRNA expression after PASMCs were exposed to hypoxia for 12 and 24 h respectively. And their protein expressions were determined by Western blotting and immunocytochemical method.Results The mRNA and protein expressions of SM-α-actin and PKGIa could be detected in PASMCs exposed to hypoxia or not, of longer exposure or shorter. The levels of mRNA and protein of PKGIa and SM-α-actin were decreased after PASMCs were exposed to hypoxia, suggesting a relationship of their expressions with hypoxia.Conclusion PKGIa signaling pathway might play an important role in the changes of cell phenotype induced by hypoxia.
