山苦茶提取物对动脉粥样硬化防治作用的研究

摘　　要：目的:探讨山苦茶(mallotus furetianus)提取物B对动脉粥样硬化(atherosclerosis,AS)的防治作用及其机制。方法:一次性给大鼠腹腔注射VD370万U/kg,高脂饮食,建立AS模型,并预防性给予山苦茶B高剂量(0.081 g/kg)、低剂量(0.026 g/kg)处理。9周后,取血测定TG、TC、LDL-C、HDL-C、SOD、NOS、MDA水平;引流胆汁,测胆汁中TB和胆固醇;取肝脏,称肝湿重,计算肝指数;迅速取胸主动脉,制成宽3 mm的胸主动脉环,保留内膜,并测定其内皮依赖性舒张能力,余HE染色,进行病理检查。结果:山苦茶B降低AS大鼠血清MDA、TC、TG、LDL-C水平,提高NOS,SOD活性;增加HDL-C/LDL-C,增加胆汁中TB的排出,改善血管组织病理变化,增强内皮依赖性血管舒张能力。高剂量山苦茶B能降低AS大鼠的肝指数。结论:山苦茶B可以防治动脉粥样硬化,这可能与其降低TC、增强NOS活性、清除脂质过氧化产物、提高机体抗氧化能力有关。atherosclerosis （AS） kg one time, dose （0.026 feeding Objective： To determine the extract B of mallotus furetianus on the prevention and therapy of and its mechanism. Methods： AS models were established by injecting rats VD370 000 U / high - fat diet, and giving extract B of mallotus furetianus high - dose （0.081 g/kg）, low - g/kg） respectively. TG, TC, LDL- C, HDL- C, SOD, NOS, MDA level of the blood were assayed in nine weeks; TB and cholesterol in the bile were measured from drainage of bile; the livers of the rates were removed and weighed, the liver index was calculated; thoracic aortas were removed instantly, thoracic aorta rings of 3 mm in width were made, the endothehum was reserved and the capacity of endothelium - derived relaxing was de- termined, the rest of the thoracic aortas were undergone HE staining and received pathological examination. Results： Extract B of maUotus furetianus lowered MDA, TC, TG, LDL - C levels of AS serum, elevated NOS, SOD activity, increased HDL - C/LDL - C, increased discharge of TB from bile, improved pathological changes of vascular tissue, increased the ability of vaso＇dilation of the endothelium - dependence. High - dose extract B of mallotus furetianus decreased the index of the liver. Conclusion： Extract B of maUotus furetianus may prevent athero- sclerosis, which may be related to the decrease of TC, enhancement of NOS activity, removal of lipid peroxidation, improvement of antioxidation.
