Optimization of Extraction Process of Paeoniae Radix and Salvia Miltiorrhiza in Chidan Pills by Uniform Design Method

摘  要:采用均匀设计法优化了赤丹丸中赤芍和丹参的提取工艺。以芍药苷和丹参酮ⅡA的含量为指标,考察了乙醇体积分数、乙醇用量、提取时间对提取效果的影响。结果表明,对芍药苷的提取,提取时间影响较大,乙醇体积分数有一定的影响,提取时间与乙醇用量有交互作用;对丹参酮ⅡA的提取,提取时间影响不大,乙醇用量有一定的影响。确定赤芍和丹参的最佳提取工艺为:6倍量的95%乙醇回流提取30 min。为赤丹丸及赤芍、丹参单味药的提取工艺提供了依据。The extraction process of Paeoniae radix and Salvia miltiorrhiza in Chidan pills were optimized by uniform design method. With the content of peoniflorin and tanshinone Ⅱ A as the determination indexes and HPLC as the examination method, the effects of ethanol volume fraction, ethanol amount and extraction time were studied. The results showed that, for extraction of peoniflorin,the influence of extraction time was significant and ethanol volume fraction had some effects on it, and there was interaction existed between extraction time and ethanol amount;for extraction of tanshinone Ⅱ A, extraction time didn't affect it much and ethanol amount had some effects on it. The optimum extraction process for peoniflorin and tanshinone It a was with 6 BV 95 % ethanol for reluxing 30 min. It provided reference to extraction processes of Chidan pills, Paeoniae radiz and Salvia miltiorrhiza.
