摘  要:目的探讨苦参素对活化肝星状细胞(HSC)整合素αv的表达和矢巢凋亡的影响。方法把培养激活的大鼠肝星状细胞株(HSC—T6)分为实验组和对照组,实验组以不同浓度的苦参素干预,蛋白质印迹(Western Blotting)法检测整合素αv的表达,末端脱氧核苷酸转移酶介导的脱氧三磷酸尿苷缺口末端标记法(TUNUL)检测凋亡指数。结果药物干预后整合素αv的表达随干预浓度的增加而逐渐降低(P〈0.05).凋亡指数则逐渐增高(P〈0.05),且呈浓度依赖性(r分别为-0.76、0.81);整合素αv与凋亡之间呈负相关,r为-0.79。结论苦参素能通过抑制活化HSC的整合素αv的表达促进其凋亡,在肝纤维化治疗中具有重要意义。Objective To explore the effect of Kwoninone on integrin αv Expression and anoikis in activated hepatic stellate cells. Methods HSC T6 was incubated and divided into control group and study group. Western Blotting and TUNUL were employed to estimate the Expression of integrin αv and apoptosis of HSC T6. Results When kwoninone was used, the expression of integrin αv significantly decreased in study group, compared with control group ( P 〈 0.05 ), and the apoptosis index was higher after treatment with kwoninone. There was a significantly negative correlation between integrin αv and anoikis (r = - 0.79). Conclusion Kwoninone can induce the apoptosis of HSCT6 by inhibiting expression of integrin αv.
