摘  要:目的:了解脂联素基因+45T/G多态性在原发性高血压(EH)患者中的分布,探讨该多态性与血浆脂联素浓度的相关性。方法:采用聚合酶链式反应-限制性内切酶长度多态性(PCR-RFLP)技术,对151例原发性高血压患者和100例正常对照者脂联素基因+45T/G多态性进行基因分型。采用ELISA法测定血浆脂联素浓度。结果:(1)原发性高血压组G/G基因型频率明显高于正常对照组(P<0.01),G等位基因频率也明显高于正常对照组(P<0.01),T等位基因频率明显低于正常对照组(P<0.01)。(2)G/G+T/G基因型的血浆脂联素浓度与T/T基因型差异有统计学意义(P<0.01)(。3)原发性高血压组血浆脂联素浓度低于正常对照组,差异有统计学意义(P<0.01)。结论:脂联素基因+45T/G多态性、血浆脂联素浓度与高血压有相关性,提示脂联素基因可能是原发性高血压的一个新易感基因。Objective: To investigate the genotype distribution of adiponectin + 45 nucleotide T/G polymorphism in essential hypertension patients, and study the relationship between polymorphism and plasma adiponectin level. Methods: One hundred and fifty-one patients with essential hypertension and 100 health subjects as normal control were recruited in this study. All subjects were genotyped for the polymorphism of adiponectin by PCR-RFLP assay. Plasma adiponectin level was determined by ELISA. Results: ( 1 )The frequencies of G/G genotype and G allele in essential hypertension group were higher than those innormal control (P 〈 0.01 ). The frequency of T allele in essential hypertension group was lower than that in normal control (P 〈 0.01 ). (2) The plasma adiponectin level in essential hypertension was lower than that in normal control (P 〈 0.01 ). (3)The plasma adiponectin of G/G+T/G genotype were lower than that of T/T genotype (P 〈 0.01 ). Conclusion: The polymorphism of adiponectin +45 nucleotide T/G is associated with plasma adiponectin level, and the adiponectin gene may be a new susceptible gene to essential hypertension.
