中药干预对剥夺性豚鼠近视动物模型的实验研究

摘　　要：目的:观察在中医理论指导下应用中药方剂对小样本实验性豚鼠近视眼的影响。方法:将15只豚鼠随机分为正常对照组(Ⅰ组)3只、单纯近视诱导组(Ⅱ组)5只、中药喂养近视诱导组(Ⅲ组)7只。Ⅱ、Ⅲ组缝合豚鼠单眼诱导形觉剥夺性近视眼模型;A超测量眼球各项指标、免疫组织化学荧光染色和Western blot印迹法检测基质金属蛋白酶-2在视网膜中的含量。结果:三组间双眼前房深度(AC),晶体厚度(L),玻璃体腔深度(V)及眼轴长度(AL)值比较,右眼(近视诱导眼)V及AL值有显著性差异;三组豚鼠右眼V及AL值两两比较发现,各组间均有显著性差异。结论:中药干预可减轻豚鼠形觉剥夺性近视的进展,其途径可能是直接或间接诱导视网膜色素上皮-脉络膜层MMP-2表达减少,从而抑制或减少了巩膜细胞外基质降解,中医阴阳辨证理论对近视眼的研究有一定指导意义。Objective：To observe the impacts of Chinese herbal formulae on the experimental myopia of small samples in guinea pig model in the guidance of Chinese medicine theory. Methods：15 guinea pigs were randomized into normal control group（group Ⅰ, 3 cases）, simple myopia induction group（group Ⅱ ,5 cases）,myopia induction group of Chinese herb breeding （ Ⅲ group,7 cases）. In groups Ⅱ and Ⅲ ,the unilateral eyes of guinea pigs were sutured to induce form-deprivation myopia. Uhrasonometry A was used to measure every index of eyeball, and immunohistochemistry and immunofluorescence as well as western blot method were adopted to detect the expression of matrix metalloproteinase-2 （MMP-2）in the retina. Results：Significant difference was presented in anterior chamber depth,lens thickness,vitreous depth and axial length of bilateral eyes and in vitreous depth and axial length in the fight eyes（myopia induction eyes）by comparison among three groups. Significant differences were presented in vitreous depth and axial length of the right eyes in three groups in paired comparison. Conclusion：The intervention of Chinese herbal formula postpones the development of form-deprivation myopia in the guinea pigs. Probably,its mechanism is relevant directly or indirectly to the reduced expression of MMP-2 in the retinal pigment epithelium-choroid layer so that the extracellular matrix degradation is inhibited or reduced in the sclera. Differentiation of syndrome in light of yin-yang theory in Chinese medicine provides certain guidance in the study on myopia.
