疏肝益肾解郁方对抑郁雌鼠性激素及神经递质的干预作用研究

摘　　要：目的:观察疏肝益肾解郁方应用于慢性不可预见应激模型的雌性抑郁症大鼠的影响,研究大鼠的行为学、性激素、脑皮层单胺类神经递质的变化。方法:将敞箱实验得分结果在符合标准范围内的72只雌性大鼠随机分为正常组、模型组、西药组、中药组,共4组,每组18只。模型组、西药组、中药组大鼠进行慢性不可预见应激造模,同时西药组服用帕罗西汀,中药组服用疏肝益肾解郁方治疗,运用行为学测定和糖水消耗实验,观察疗效对比,并测定雌二醇(E2)、孕激素(P)、催乳素(PRL)和5-羟色胺(5-HT)、多巴胺(DA)的含量。结果:中药组、西药组治疗后抑郁大鼠抑郁行为改变明显,Open-Field评分及糖水消耗量与模型组比较有显著性差异(P<0.05,P<0.01),西药组与中药组比较无显著性差异(P>0.05)。中药组与抑郁相关的指标E2、5-HT、DA含量显著提高。结论:疏肝益肾解郁方对抑郁雌鼠雌激素和单胺类神经递质5-HT、DA有明显的干预作用。Objective：To study effect of Shugan Yishen Jieyu Decoction （SGJYD）on the the behavior,sex hormone,brain cortex monoamine neurotransmitter of depressed femate rats induced by chronic unpredictable stress. Methods： 72 depressed female rats selected according to the scores of open field test were devided into normal group,model group,western medieine group （with Paroxetine）and traditional Chinese medieine group （with SGJYD）by 18 rats in each group. Rats in the model, western,TCM groups were induced by ehronic unpredictable stress. The therapeutic effect was observed through behaviour test and sucrose consumption. The Ez,P,PRL, 5-HT and DA were detected. Result：The depressed behavior of rats improved greatly in TCM and western medicine groups after treatment. Open-field scores and the sucrose consumption of TCM and western groups were siginifieantty different from those of model group（P〈0.05 ,P〈0.01 ）after treatment. There were no significant difference between TCM group and western medicine group（P〉0.05）. Ez,5-HT and DA of TCM group increased remarkably. Conclusion：SGJYD is effective on sex hormone, monoamine neurotransmitter,5- HT and DA of female depressed rats.
