摘 要:目的:观察圣地红景天对1型糖尿病大鼠尿微白蛋白排泄率及肾脏TGF-β1表达的影响并探索其机制。方法:SD雄性大鼠45只,单侧肾切除后随机分为:对照组15只(NC组),模型组30只,后者按55mg/kg体重单次腹腔注射1%链脲佐菌素(STZ)制作糖尿病模型。待造模成功后随机分为糖尿病模型组(DC组)、糖尿病治疗组(DR组)各15只。分别于第4、8、12周时检测各组血尿素氮、肌酐及尿肌酐、尿微白蛋白排泄率,同时测肾组织中丙二醛(MDA)含量,铜锌超氧化物歧化酶(Cu-Zn-SOD)及谷胱甘肽过氧化酶(GSH-Px)活性,留取肾脏组织分别作光镜、电镜检查及免疫组化检查TGF-β1的表达。结果:与NC组相比,DR组24h尿微白蛋白排泄率下降,肾组织中NDA含量下降,Cu-Zn-SOD及GSH-Px活性升高;与DC组相比,DR组尿蛋白排泄率减少,早期肾小球高滤过减轻,病理检查示系膜增生明显减轻,电镜示肾小球毛细血管基底膜增厚轻于DC组,免疫组化分析示DR组TGF-β1表达较DC组明显减少。结论:圣地红景天可以有效减轻糖尿病大鼠的肾脏损害,其对糖尿病肾脏的保护作用可能与其抗氧化应激及降低肾组织TGF-β1的表达有关。Objective: To investigate the effect of rhodiola .sacra on micro - albumin excretion rate and the expression of TGF- -β1 in kidney of type 1 diabetic rats. Methods:Fourty - five male SD rats with unilateral kidney removed were randomly divided into the control group(NC group, n = 15) and the diabetic model group( n = 30). Diabetic rats models that were established by single peritoneal injection of 1% STZ (55 mg/kg) were subdivided into the diabetic group (DC group, n - 15) and the diabetes treatment group(DR group, n - 15). We detected the serum BUN, serum creatinine, urine creatinine,micro albumin excretion rate in each group at the 4th, 8th and 12th week respectively. The MDA level, Cu, Zn SOD and GSH - Px activity in kidney tissue were examined, pathology and ultrmierostructure and the expression of TGF --β1 were examined by microscope, electron miercxscope and immunohistoc hemistry respectively. Results:Compared with those in the NC group, micro- albumin excretion rate and the concentration of MDA in the kidney were significantly decreased in the DR group, while Cu, Zn - SOD and GSH - Px activity in kidney tissue were significantly increased in the DR group. Compared with those in the DC group, the micro albumin excretion rate was decreased;glomeruli high filtration and mesangial hyperplasia relieved and the degree of basal membrane thickening and the expression of TGF-β1 were significantly decreased in the DR group. Conclusion:Rhodiola sacra can effectively relieve the damage of kidney in the diabetic rats, its protective effect may be related to the decrease of oxidative stress and expression of TGF-β1.
