复方蒲黄颗粒对大鼠脑缺血再灌注后GFAP表达的影响

摘　　要：目的研究复方蒲黄颗粒对大鼠脑缺血再灌注后胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的影响。方法实验采用90只清洁级SD大鼠,随机分为6组(每组15只):假手术组(Sham组),模型组(I/R组),复方蒲黄大剂量组(PHH组),复方蒲黄中剂量组(PHM组),复方蒲黄小剂量组(PHL组),脑血栓片组(NXS组)。采用右侧颈总动脉夹闭手术,建立大鼠脑缺血再灌注模型,分别在术后6 h、24 h、72 h,运用TTC染色计算脑梗死体积,运用免疫组化方法检测脑组织GFAP的动态变化。结果GFAP的表达呈现随时间增加而增长,与I/R组相比较,PHH组、PHM组在术后24 h、72 h表达增长显著增加(P<0.05)。结论复方蒲黄颗粒能够有效减小大鼠脑缺血再灌注后脑梗死体积,调节脑缺血再灌注损伤后GFAP蛋白表达,以大、中剂量作用最为显著,这可能是本方对全脑保护的重要疗效机制之一。Objective To study the effect of Pu-huang Recipe on the expression of glial fibrillary acidic protein （GFAP） after cerebral ischemia/reperfusion injury in rats Methods Ninety Sprague-Dawley rats were divided into six groups randomly （ 15 rats in each group） ： sham group, cerebral ischemia reperfusion group（ I/R group）, high dose Pu-huang Recipe group （PHH group）, medium dose Pu-huang Recipe group （PHM group ）, low dose Pu-huang Recipe group （PHL group）, Nao-xueshuan troche group （NXS group）. The models of cerebral ischemia/reperfusion injury were established by right common carotid artery blocking. The volume of cerebral infraction were detected by TTC staining at 6 h ,24 h ,72 h after reperfusion. The dynamic changes of GFAP in cerebral tissue were investigated by using immunohistochemiscal technique.Results The expression of GFAP was increased as the time went by. Compared with I/R group, the expressions of GFAP increased significantly in PHH and PHM group at 24 h and 72 h after operation （ P 〈 0.05）. Conclusion Pu-huang Recipe especially in high dose and medium dose have protective action on cerebral infarction and can decrease the expression of GFAP after cerebral ischemia/reperfusion injury in rats. This may be one of the mechanisms of Pu-huang Recipe on protection of brain.
