摘 要:目的:研究表皮生长因子(EGF)和胰岛素样生长因子-1(IGF-1)体外诱导胎儿骨髓间充质干细胞(BMSCs)向上皮细胞的分化,及其对种植于羊膜上BMSCs的影响。方法:不同浓度EGF、IGF-1体外诱导第3代BMSCs培养7 d后,检测各组细胞角蛋白(CK)及上皮膜抗原(EMA)的阳性细胞率。检测去上皮羊膜有无EGF或IGF-1表达情况;观察生长因子对种植于羊膜上BMSCs的分化影响。结果:EGF 30 ng/ml或IGF-1 50 ng/ml组的CK及EMA阳性细胞率高于其余剂量组;两因子联合诱导组的CK及EMA阳性细胞率均高于各单因子组;30 ng/ml EGF和50 ng/ml IGF-1联合组的阳性细胞率最高;去上皮羊膜表达EGF,不表达IGF-1;BMSCs在羊膜上生长不受外源性生长因子影响。结论:EGF和IGF-1在体外均有诱导BMSCs向上皮细胞分化的作用;以30 ng/ml EGF和50 ng/ml IGF-1联合组的协同作用最强;种植于羊膜上的BMSCs可生长为复层上皮结构,羊膜中的EGF可能是其诱导作用的内源性因素。Objective: To study the differentiation of fetal bone marrow mesenchymal stem cells (BMSCs) induced by epidermal growth factor (EGF) and insulin-like growth factor-1 (IGF-1) in vitro and these effects on the BMSCs planted on amnion. Methods: The positive rate of cytokeratin (CK) and epithelial membrane antigen (EMA) of cells was detected in each group at 7 clays after P3-BMSCs being induced by different concentrations of EGF and IGF-1. Whether EGF and IGF1 was expressed in the amniotic membrane without epithelium was detected, and the differentiation of BMSCs planted on amnion with or without growth factors was observed. Results : The positive cells of CK or EMA in the group of EGF 30 ng/ml or IGF-1 50 ng/ml was significantly higher than those of other concentration groups; The CK or EMA positive cells in the group induced with EGF and IGF-1 were significantly higher than those in the group induced with single factor; The CK positive cells in the group induced with 30 ng/ml EGF and 50 ng/ml IGF-1 was higher than those in other groups induced with two factors; EGF was expressed in the acellular amniotic membrane, but IGF-1 was not expressed. BMSCs formed stratified epithelium on the amnion with or without the exogenous growth factors. Conclusion: BMSCs could be induced to differentiate into epithelial cells by EGF and IGF-1 in vitro ; The optimal synergistic effect of EGF and IGF1 was in 30 ng/ml EGF and 50 ng/ml IGF-1 respectively. The BMSCs planted on the amnion could grow and form stratified epithelium structure. EGF in amnion may be one of the endogenetic reasons which induced BMSCs differentiate into epithelial cells.
