摘 要:目的:观察骨灵片对原发性骨质疏松症(POP)大鼠骨密度(BMD)及血清OPG、RANKL表达的影响。方法:将大鼠随机分为正常组、模型组、阳性对照组、骨灵片高、中、低剂量组,采用去势法建立大鼠POP动物模型,正常饲养1月后,骨灵片3个剂量组和阳性对照组分别给予骨灵片和护骨胶囊治疗3个月后处死,采用X线骨密度仪和ELISA法分别测定大鼠股骨BMD及血清OPG、RANKL。结果:与模型组比较,各给药组BMD值均较高,其中骨灵片高、中剂量组BMD与模型组比较有显著性差异(P<0.05),骨灵片3个剂量组及阳性对照组与正常组比较无显著性差异(P>0.05)。骨灵片3个剂量组、阳性对照组及模型组的血清OPG和RANKL均较正常组升高,而骨灵片3个剂量组、阳性对照组测得较高的OPG和RANKL值,较之模型组有显著性差异(P<0.05,P<0.01)。结论:骨灵片可通过上调OPG,下调RANKL的表达,影响OPG/RANKL/RANK系统,抑制骨丢失,维持BMD,从而起到防治POP的作用。Objective:To investigate the effect of Gu Ling Pian(GLP) on bone mineral density(BMD) and the expressions of serum OPG and RANKL in primary osteoporosis(POP)rat models. Methods: Rats were divided randomly into sham-operated group,model group,positive control group,high dosage GLP group,middle dosage GLP group and low dosage GLP group. POP animal model was established by castration in rats. After normal feeding for one month, Gu Ling Pian and Hu Gu Jiao Nang were administrated in three GLP groups and positive control group respectively and the rats in the groups were all sacrificed three months later. Bone's X-ray measuring instrument and ELISA were adopted to measure femoral BMD and the levels of serum OPG and RANKL respectively. Results:Comparing with model group, BMD level was quite higher in every medication group, but, BMD levels in GLP groups of high and middle dosages were different significantly in comparison with that in model group(P〈O.05). There was no significant difference in comparison of GLP groups of three dosages and positive control group with sham--operated group (P〉0.05). The levels of serum OPG and RANKL in the three GLP groups,positive control group and model group were all increased comparing with sham-operated group,of which,the levels of OPG and RANKL in three GLP groups and positive control group were higher than those in model group,indicating significant dif- ference(P〈0.05 ,P〈0.01 ). Conclusion:Gu Ling Pian up-regulates OPG,down-regulates the expression of RANKL, impacts OPG/RANKL/RANK system,inhibits bone loss and maintains BMD so that it can prevent and treat primary osteoporosis.
