摘 要:采用液-液分配萃取方法,从大豆荚壳甲醇提取物中分离获得石油醚、氯仿、乙酸乙酯、正丁醇和水的萃取物,测定了其对南方根结线虫J2的毒杀活性,结果表明正丁醇萃取物比其它4种萃取物有更高的杀虫活性,24和48h对线虫校正死亡率分别为57.76%和67.72%。将正丁醇萃取物通过硅胶柱层析和薄层层析,得到8个组分,通过杀虫活性试验,得到组分2和7为主要的杀虫活性组分,24和48h对线虫校正死亡率分别为65.93%、66.65%和69.25%、68.51%。进一步就组分2和7对J2抗氧化酶活性的影响进行了研究,结果表明两种组分对谷胱甘肽转硫酶和过氧化氢酶活性都有一定的抑制作用,其中两种组分对过氧化氢酶活性的抑制效果差异不显著,组分7对谷胱甘肽转硫酶活性具有显著的抑制效果,24和48h的抑制率分别可达72.36%和50.21%。说明大豆荚壳的杀线虫活性成分可能通过影响虫体内的氧化代谢,使线虫体内氧化和抗氧化作用失衡,最终导致线虫死亡。Five fractions of soybean hull extracts were obtained with different polarities of solvents such as petroleum ether,chloroform,ethyl acetate and n-butanol and water by the method of liquid-liquid extraction,and then their nematitidal activities were tested. The results showed that n-butanol extracts were the most nematicidal one among the five fractions, and in 24 and 48 h of treatment, the revised mortality of J2 was 57.76% and 67.72%, respectively. The chloroform extract was further separated through aluminum oxide column and the fractions were monitored with analytical thin layer chromatography (TLC). Eight fractions were obtained and also tested for nematicidal activity. The results indicated that fraction 2 and 7 had higher nematicidal one than the other fractions. The revised mortality of fraction 2 was 65.93% in 24 h of treatment and 69.25% in 48 h of treatment,and that of 7 was 66.65% in 24 h of treatment and 68.51% in 48 h of treatment. In addition,effects of the two fractions on catalase (CAT) and glutathione S-transferase (GST) of J2 were studied. The results showed that the activities of both enzymes were inhibited. The difference of inhibiting effect on catalase between them was not significant,the inhibiting effect on glutathione S-transferase of fraction 7 was higher than that of 2 significantly,and in 24 and 48 h of treatment,the inhibiting rate could be up to 72.36% and 50.21% ,respectively. It is concluded that the nematicidal components from soybean hull are likely to destroy the balance of oxidation and anti-oxidation by affecting the oxide metabolism and in the end result in death of J2.
