摘  要:以鲫鱼(Carassius auratus)为试验材料,研究了在离体条件下经不同质量浓度的十溴联苯醚(PBDE-209)暴露后,鲫鱼肝脏线粒体中总抗氧化能力(T-AOC)、丙二醛(MDA)含量、黄嘌呤氧化酶(XOD)和超氧化歧化酶(SOD)活性的动态变化。结果表明,采用质量浓度为5.6-100.0 mg/L的PBDE-209处理鲫鱼肝脏线粒体30 min,5.6 mg/L组鲫鱼肝脏线粒体中T-AOC、MDA含量、XOD和SOD活性与对照组相比无显著差异(p〉0.05);其余各组的XOD活性和MDA含量随PBDE-209质量浓度增加逐渐上升,而T-AOC和SOD活性逐渐下降,均与PBDE-209质量浓度呈明显的相关关系(p〈0.01)。这说明PBDE-209对鲫鱼肝脏产生了氧化损伤,具有生化毒性。离体条件下鲫鱼肝脏线粒体中的T-AOC、MDA、XOD和SOD可作为生物标志物来评价多溴联苯醚的生化毒性。The present paper intends to introduce the authors' experimental research on the toxic effects of decabromodiphenyl ether (PBDE- 209) on the content of maleic dialdehyde (MDA), the total anti-oxidative capacity (T - AOC) and the activities of xanthine oxidase (XOD) and superoxide dismutase (SOD) in mitochondria isolated from the liver of Carassitas auratus which exposed to PBDE -209. For the research purpose, we have isolated the mitochondria from the liver of the fishes to be tested and put them back to water with the concentration of PBDE - 209 being, 0 mg/L, 5.6 mg/L, 10.0 mg/L, 18.0 mg/L, 32.0 mg/L, 56.0 mg/L and 100.0 mg/L. The said four indicators were then analyzed after a 30 min exposure in vitro at the temperature of 25℃. Just as the normal values of XOD and SOD would do (6.64±0.15) U/g prot and (12.50±0.25) U/rag prot, the results of our experiments prove that the normal values of MDA and T - AOC tend to be separately (5.75 ± 0. 10) nmol/mg prot and (3.09 ± 0.09) U/mg prot. And the test group of 5.6 mg/L PBDE 209 has no effects on T - AOC, MDA, XOD and SOD in mitochondria after treated 30 min( p 〉 0.05). But the other test groups can cause the obvious changes to T- AOC, MDA, XOD and SOD (p 〈 0.01). With the concentration of PBDE- 209 increased, MDA, XOD tends to increase while T - AOC, SOD are lowering, which turns to prove T- AOC, XOD, MDA, SOD and PBDE -209 concentrations are closely related. The above results can clearly indicate that the oxidative stress and the biochemical toxicity to the mitochondria isolated from the liver of Carassius auratus with PBDE - 209 which is above 5.6 mg/L. T- AOC, MDA, XOD and SOD in liver mitochondria of fishes can be taken as a biomarker to assess the biochemical toxicity of polybrominated diphenyl ethers.
