摘 要:[目的]为进一步了解家蚕酚氧化酶原基因的功能奠定基础。[方法]根据GenBank上登录的家蚕酚氧化酶原基因(PPO1和PPO2)的cDNA序列设计2对特异引物,通过半定量RT-PCR技术检测家蚕幼虫各发育时期(蚁蚕到5龄起蚕)酚氧化酶原基因的转录活性。[结果]PPO1和PPO2基因在家蚕幼虫各发育时期的转录活性存在较大差异。PPO1基因在2龄眠蚕、4龄眠中的表达量最高,其次是2龄起蚕、3龄眠蚕3、龄起蚕和1龄眠蚕,在3龄眠中、4龄起蚕的表达量较低,在4龄眠蚕有弱表达,在其他发育时期几乎无表达。PPO2基因除了在蚁蚕1、龄眠中、2龄眠蚕无表达外,在其他时期的表达量均较高。[结论]酚氧化酶原基因在家蚕幼虫各发育时期的转录表达均具有明显的时空特异性。[ Objective ] The aim was to lay the foundation for further understanding the function of prophenoloxidase gene from Bombyx mori. [ Method] 2 pairs of specific primers were designed according to the cDNA sequences of pmpbenoloxidase genes ( PPOland PPO2) from B. mori reported in the GenBank and the transcriptional activity of pmphenoloxidase gene in every developmental stages (from newly hatched larva to sillkworm in the beginning stage of the 5^th-instar melting of B. mori larva were detected by semi-quantitative RT-PCR technique. [ Result] There was large difference among the transcriptional activities of PPO1 and PPO2 gene in every developmental stages of B. mori larva. The expression of PPO1 gene in the 2^nd instar molting silkworm and in the middle of 4th instar molting silkworm were highest, followed by the 2nd instar moulted silkworm, 3rd instar melting silkworm, 3rd instar moulted silkworm and 1st instar melting silkworm, that in the middle of 3rd instar molting silkworm and 4th instar moulted silkworm was lower. There had weak expression in the 4th instar melting silkworm and there had no expression in other developmental stages. PPO2 gene was not expressed in the newly hatched larva, the middle of 1st instar molting silkworm and the 2nd instar melting silkworm and it had higher expression in other developmental stages. [ Conclusion] The transcriptional expression of prophenoloxidase gene in every developmental stages of B. mori larva had obvious specificity of time and space.
