摘 要:目的对幽门螺杆菌(HP)HPAG1_1148基因进行生物信息学分析,为HP疫苗和细菌学研究奠定基础。方法应用Omiga2.0等生物软件和Swiss—port、GenBank等数据库分析HPAG1_1148基因的变异性及其表达蛋白的化学性质、分子结构、保守区、抗原结构域、抗原表位。结果该基因同源性为93.7%-97.4%,表达蛋白N端有长约23aa的信号肽,为分泌型蛋白,具有抗原结构域、B细胞抗原表位及与胞膜孔道蛋白相似的保守区。结论该基因具有较高保守性,表达蛋白是一种分泌型外膜蛋白,可能具有转运功能和抗原分子结构特征,有望成为HP疫苗。Objective To investigate variability of the helicobacter pylori gene HPAG1 1148 for vaccine development and bacteriological research. Methods The bioinformatics software Omiga 2.0 and Swiss-port, GenBank data were employed to analyze variability of the gene and to predict chemical profiles, structure, conservative domains, antigenicity of the encoded protein. Results The gene was 93.7% - 97.4% in conservation. There was a 23 amino acids long signal peptide at the N-end of the putative peptide, which suggested as a secretary protein, with antigenic domains, B-ceU epitopes, a conservative domain similar to porin also putatively being in the protein. Conclusions The HPAG1 1148 gene is rather conservative and can encode outer membrane protein, which may show roles in membrane's transportation and molecular structural profiles of antigens, thus valuable for the development of HP vaccine.
