寿光鸡MSTN基因3种不同转录子的克隆及序列分析

摘　　要：采集1日龄寿光鸡腿肌,提取RNA进行RT-PCR反应、TA克隆和测序,序列测定结果表明:寿光鸡腿肌中存在3种不同大小的MSTN cDNA。第1种MSTN cDNA由1128bp组成,将该序列命名为csMSTN-1,与已报道的鸡MSTN cDNA序列(gb:AF019621.1)同源性为99%;第2种MSTN cDNA由985bp组成,命名为csMSTN-2,与csMSTN-1相比,缺失了374～517处的143个碱基,并且在6处存在碱基变化;第3种MSTN cDNA由754bp组成,命名为csMSTN-3,与csMSTN-1相比,缺失了374～748处的374个碱基,并且存在26处碱基变化。这是世界上首次发现鸡体内存在3种不同大小的MSTN基因转录子,该结果为进一步确定MSTN基因的作用机理提供了新的试验依据。RT-PCR was applied in this experience to study the expression of myostatin. It was found that there were three MSTN gene transcriptons of varied sizes in leg muscle of shouguang chicken aged 1 day. By cloning and sequencing, in the results showed that, one MSTN cDNA was compcsed of 1128 bases, and was named as cdMSTN-1, which held 99% homology with the chicken MSTN cDNA reported （gb： AF019621.1）.Another MSTN cDNA consisted of 985 bases, named as cdMSTN-2,143 cases from 374 to 517 were deleted compared to that of cdMSTN-1, and six bases were changed. The third MSTN cDNA was composed of 754 bases, and 374 cases from 374 to 748 were deleted compared to that of cdMSTN-1, was named as cdMSTN-3. The results provided experimental basis to study the mechanism of MSTN gene.
